Vaccine and adjuvant response profiling in human immune organoids
Animal studies do not always capture human vaccine responses. Human immune organoids offer an additional way to study antigen-specific B cell responses before clinical testing.
What studies can compare
Studies focus on comparative human B cell responses, with objectives agreed in advance.
Antigen designs
Compare antigen constructs, such as different stabilized forms of the same protein, for the B cell responses each produces.
Adjuvants
Compare the same antigen with different adjuvants under matched conditions. We discuss during design which cells and mechanisms the model represents, since performance in culture can differ from performance after administration.
Donor variation
Where donor availability and characterization allow, compare responses across donors, including donors of different ages.
A study, step by step
Study design
We agree on the antigens, adjuvants, comparators, and readouts, and the decision the study needs to inform.
Donor panel
We select donors to fit the question, subject to donor availability and characterization.
Organoid exposure
Donor immune cells are cultured in organoids and exposed to each condition in parallel.
Readouts and report
We measure the agreed readouts, such as antigen-specific B cell activation and antibody production, and report results by condition and donor.
Organoid studies are preclinical and provide earlier information to support candidate selection. Clinical studies remain necessary, and organoid results do not replace in vivo studies or clinical trials.
Questions
Who runs the work?
Projects are designed and coordinated by the OrganoAb team, and we describe the operating plan for each project during scoping.
Can you test our own adjuvant?
We can discuss partner-supplied materials, including whether the relevant mechanisms are represented in the model, during study design.
Can we discuss a project confidentially?
Yes. We put a confidentiality agreement in place before any detailed discussion of your molecules or targets.
